Elranatamab in Refractory Multiple Myeloma with Prior BCMA Therapies
Evaluating the safety and efficacy of elranatamab
Dr. Ajay Nooka presents the efficacy and safety analysis of a drug called elranatamab in patients with refractory multiple myeloma who had prior exposure to BCMA-directed therapies. Elranatamab is a humanized bispecific antibody that targets both BCMA-expressing myeloma cells and silica-expressing T cells.
The pooled analysis included data from the ongoing phase 2 MAGNITUDE-3 study and evaluated the monotherapy of elranatamab in BCM-naive patients with relapsed/refractory myeloma. The analysis included a total of 87 patients, of which 59 received a prior antibody-drug conjugate (ADC) and 36 received a prior CAR-T therapy. The median age of patients in the ADC cohort was younger, and high-risk cytogenetics were observed in 30% of the patients. The median lines of therapy were seven overall. The study showed an overall response rate of 61% and a median duration of response that has not been reached yet. The safety profile of elranatamab was consistent with that of BCM-naive patients.
