DREAMM-7 update: Subgroup analyses from a phase 3 trial of belantamab mafodotin (belamaf) + bortezomib and dexamethasone (BVd) vs daratumumab, bortezomib, and dexamethasone (DVd) in relapsed/refractory multiple myeloma (RRMM)
Summary:
In patients with relapsed/refractory multiple myeloma (RRMM) and poor prognostic features, there is a significant unmet need for effective treatments. The DREAMM-7 trial (NCT04246047) evaluated the efficacy of belantamab mafodotin (BVd) versus standard-of-care daratumumab plus bortezomib and dexamethasone (DVd) in RRMM patients with at least one prior line of treatment. The study demonstrated a statistically significant and clinically meaningful progression-free survival (PFS) benefit with BVd compared to DVd. Further analyses from DREAMM-7 explored efficacy in key subgroups, including patients refractory to lenalidomide and those with high-risk cytogenetics. BVd showed superior PFS and overall response rates (ORR) compared to DVd in these subgroups. Ocular adverse events were more frequent with BVd, but overall, the safety profile was manageable. The results support BVd as a potential new standard of care in RRMM patients with high unmet needs.
